Enhancing intracellular delivery of a broad range of proteins

DIVTECH Protein Kit, based on reactive lipids, are suitable for click chemistry to conjugate any azide-modified protein. The proteins need to be functionalized with a linker (azide-dPEG8-NHS ester) for an efficient association to DIVTECH by following easy and mild procedures that do not compromise its structure and activity. The azido modified protein will covalently react with the surface of the DIVTECH formulation. The linker for the protein modification is provided within the kit.

Intracellular delivery of RLuc8 protein to L3.6pl cells using DIVTECH. RLuc8 activity was measured after protein activation and detected by a luminometer. Signal was only detected (455 nm) after using DIVTECH Protein Kit, showing that the protein was efficiently delivered.
DIV031
Intracellular delivery of the enzyme β-Galactosidase to L3.6pl cells using DIVTECH. The β-Galactosidase activity (blue signal) was significantly enhanced when delivered by DIVTECH and showed no toxic effects as compared to commercially available competitors where cell debris and changes in the cell morphology were observed.
